DNA electrophoresis is laboratory technique used for separation and visualization of DNA sample, through which qualitative and semi-quantitative characterization of DNA can be achieved. DNA size in reference to the standard DNA ladder, the presence of unwanted DNA / RNA fragments can be determined using electrophoresis technique. The procedure involves applying electric current to agarose gel loaded with DNA sample. To facilitate the flow of current and DNA fragments through the gel, the gel is immersed in a buffer such as Tris AcetateEDTA (TAE). Buffers like TAE have a low buffering capacity, but provide the best resolution for larger DNA and is generally the preferred choice. The migration of DNA in the gel is then visualized under UV, wherein it can be seen in the form of bands at different gelpositions dependent on the fragment size and charge.
