Some polymerase enzyme didnt show proper amplification as they can not bind properly. This can be avoided by adding an enzyme after the initial denaturation. The enzyme remains inactive until the reaction mixture reaches higher temperature. It should be stored at -20-degree Celsius for long term use and activity of an enzyme. Avoid frequent freeze and thaw to maintain the stability of polymerase.
