GeNei Instant Cloning Kit is a system which is provided to make cloning more convenient. It is known that PCR amplification by Taq DNA polymerase, and other thermostable polymerases generates a product which has a 3 A. Since this A is added in a temple-independent manner, it causes a lot of potential problems during cloning. The GeNei Instant Cloning Kit offers a solution to this problem by providing a T-vector. This is a special type of vector which has a 3 T overhang and hence, is able to solve the problem of the 3 A overhang in the PCR product.

The kit contains the following features and components:

(a) INSTANT T4 DNA ligase and 2X INSTANT ligation buffer these components are provided to enhance the otherwise slow process of single base overhang ligation.

(b) The ligation can be carried out at room temperature and is expected to be complete in an hour.

(c) The kit may be used for raw or purified PCR products. The product need not be treated with enzymes.

(d) The vector contains a marker for blue-white selection, thus allowing efficient screening.

(e) The design of the vector allows for easy removal of the insert using Nco I restriction enzyme. Primers for the SP6 and T7 promoters can be used for insert sequencing.

(f) About 30-60% clones can be expected.
