The technique uses a thermostable DNA polymerase, primers and dNTPs to amplify a DNA fragment in a series of temperature-controlled steps. Nested PCR is a variation of the conventional PCR which employs two PCR reactions for the same DNA fragment to increase the amplification of specific sequences and prevent any background amplification. This is achieved by using two sets of primers, where the PCR product from one primer is again amplified using the second primer set, which targets a region within that PCR product itself.
