It is available in powder form (dark green) soluble in water, ethanol and slightly soluble in alcohol. It belongs to the phenazine group of dyes. It is also known as 8-(4-Dimethylaminophenyl)diazenyl-N, N-diethyl-10-phenylphenazin-10-ium-2-amine chloride. It was used by Leonor Michaelis in 1900 to stain mitochondria supravitally. It is also used as an indicator and changes colour relative to the concentration of oxygen. In the presence of oxygen, it oxidizes to a blue colored compound and in the absence of oxygen changes to a pink colored compound. In histology, the oxidation and reduction states show the alterations occurring in the electron transfer chain. It has also been used for staining of insect chordotonal organs and peripheral nerves, where the stained nerves appear dark blue in colour. It can be used to determine cell density by staining technique. It has the ability to stain biomolecules, chromosomes and nucleic acids and is also used to stain living organisms and tissues like fungi, brain, sperms and yeast cell. Janus Green B is also used in diagnosis of disease related to amyloid accumulation.
