It belongs to the phenazine group of dyes. It is also known as 8-(4-Dimethylaminophenyl)diazenyl-N,N-diethyl-10-phenylphenazin-10-ium-2-amine chloride. It was used by Leonor Michaelis in 1900 to stain mitochondria supravitally. It is also used as an indicator and changes colour relative to the concentration of oxygen. In the presence of oxygen, it oxidizes to a blue colored compound and in the absence of oxygen changes to a pink colored compound. In histology the oxidation and reduction state show the alterations occurring in the electron transfer chain. It has also been used for staining of insect chordotonal organs and peripheral nerves, where the stained nerves appear dark blue in colour. It can be used to determine cell density by staining technique. It has the ability to stain biomolecules, chromosomes and nucleic acids and also is used to stain living organisms and tissues like fungi, brain, sperms and yeast cell. Janus Green B is also used in diagnosis of disease related to amyloid accumulation.
