STAIN solution2 also known as Jaswant Singh and Bhattacharjee solution is 1(polychrome methylene blue). It was first synthesized in 1944 by Jaswant Singh and Bhattacharjee with 3-8 hour for dichromate-acid oxidation and addition of varying quantities of Na2HPO4 buffer for pH adjustment. It is generally composed of blue and purple components corresponding to methylene azures with methylene blue and thionin with allied products. It is widely used for the detection of malarial parasites. The parasites can be clearly visualized and both thick and thin smears can be visualized making this technique superior to Fields stain. But one drawback of this technique is that the stains fade rapidly and is therefore not preferred for permanent slide preparations. It is inexpensive, easily available and is widely used in field conditions and in government hospitals. This is the standard method used by the laboratories under the National Malaria Eradication Programme in India.
