Lysine Iron Agar Slant is supplied by TM Media. Lysine Iron Agar Slants is a type of medium used to distinguish enteric organisms based on their ability to decarboxylate or deaminate lysine and generate hydrogen sulfide. Johnson et al. employed LIA with Kligler Iron Agar to differentiate particular groups within the Enterobacteriaceae family. The medium contains a pancreatic digest of gelatin and yeast extract, which provide nitrogen, amino acids, vitamins, and other crucial elements required for bacterial growth. Bromcresol purple is used as a colour indicator to detect carbohydrate fermentation, as well as lysine decarboxylation and deamination reactions. When the pH of the medium drops below 5.2, the colour of bromocresol purple changes to yellow, and it appears purple when the pH is above 6.8. The presence of sodium thiosulfate and ferric ammonium citrate allows for hydrogen sulfide detection. Sodium thiosulfate is a substrate for enzymatic reduction, and the resultant hydrogen sulfide gas, which is colourless, reacts with ferric ammonium citrate to produce ferrous sulfide, a black insoluble precipitate that blackens the medium.
