Nickel - CL Agarose is a product by GeNei. Nickel binds with high affinity to Polyhistidine tag, or the His-tag. His-tag is essentially an amino acid of 6 or more histidine residues. In recombinant DNA technology, recombinant proteins are tagged with a polyhistidine tail and they are purified by affinity chromatography using Nickel resins. The Nickel - CL Agarose from Genei is designed for capturing his-tag fusion proteins. Using a strong chelating agent iminodiacetic acid, Ni2+ ions are crosslinked with agarose. The chelator occupies binding sites in the co-ordination sphere of nickel to allow the cross-linking, while water occupies the other 3 sites. When the histidine residues come in contact, they replace the water present in 3 sites, thus forming high affinity bonds.
