Rapid Coliform Test, also known as the Coliform Test Kit, is a method for examining water quality that detects the presence of E. coli and coliforms by analysing their enzyme-substrate reactions in water samples. The presence of coliform bacteria is used as an indicator of water quality for various purposes, such as domestic or industrial use. Enzyme substrate tests are used to detect these bacteria, whereas hydrolysable substrates are used to identify the organisms. In the enzyme technique, the total coliform group is defined as bacteria that have the enzyme -D-galactosidase, which breaks down the chromogenic substrate and releases the chromogen. E. coli bacteria are identified as those that give a positive total coliform response and have the enzyme -D-glucuronidase, which breaks down a fluorogenic substrate and releases the fluorogen.
