RNA loading dye-TRLD-100 is supplied by Transiom Genomics. RNA Loading Dye finds use in preparation of RNA ladders and RNA samples for electrophoresis on agarose or polyacrylamide gels. The loading dye contains tracking dyes like bromophenol blue and xylene cyanol FF and intercalating dye like ethidium bromide. Bromophenol blue migrates slightly faster than human 5S rRNA, whereas xylene cyanol FF migrates slightly slower than 18S rRNA as observed in most denaturing agarose gel systems. RNA Loading Dyes also contain the denaturing agent formamide, which allows the separation of RNA fragments (according to size) even in the case of non-denaturing electrophoresis. Formamide plays a role in stabilizing RNA as well. EDTA is included in the solution in order to protect sample from nuclease degradation.Loading Dye Solutions allow RNA visualization without the need for additional staining of denaturing agarose gels. But in case RNA fragments are separated on native agarose gels, additional staining with ethidium bromide is advisable. Even in the case of electrophoresis in polyacrylamide gels, the RNA ladder should be stained with ethidium bromide (0.5g/ml) for 20min, or by any other staining technique.
