Soyabean Casein Digest Agar is a general-purpose medium commonly used for cultivation of a wide variety of microorganisms. It is advisable to store this product between 20-30C. Composition (in gm/L) is as follows:
Tryptone (15.00)
Soya peptone (5.00)
Sodium chloride (5.00)
Lecithin (0.70)
Polysorbate 80 (Tween 80) (5.00)
Agar (15.00)
Glycerol (10.00)
Beta-lactamase mixture/Plate (5.00)
Soyabean Casein Digest Agar with Glycerol, polysorbate 80, Soya lecithin and beta-lactamase is used in plates for the detection and enumeration of microorganisms present on surfaces of sanitary importance and also in environmental monitoring of clean room for facilities where production of Penicillin is carried out. Various pharmacopoeias recommend using Soybean Casein Digest Agar as sterility testing medium. It is also used in validation of sterility checking procedure in accordance with the microbial limit testing harmonized methodology of USP/EP/BP/JP. This medium is used in microbial limit test and antimicrobial preservative-effective test. Casein enzymic hydrolysate and papaic digest of soyabean meal supply nitrogenous compounds and other nutrients essential for microbial replication. Lecithin and polysorbate 80 (Tween 80) are neutralizers which seem to inactivate residual disinfectants from where the sample is collected. Lecithin neutralizes quaternary ammonium compounds and polysorbate 80 neutralizes phenolic disinfectants, hexachlorophene, formalin and with lecithin ethanol. Beta-lactamase mixture added in the medium will inactivate the beta-lactam antibiotics thus enabling the growth of resistant strains present in the environment of clean rooms where production of antibiotics is carried out.
