Ssp I is a type II restriction endonuclease. Restriction endonucleases are a class of enzymes known to cleave double stranded DNA. These enzymes recognise specific palindromic sequences on the DNA, known as restriction sites. Type II restriction endonuclease search for palindromic recognition sequences on the double-stranded DNA and cleave the DNA at these specific sites. Chemically, these enzymes fragment the DNA by breaking the phosphodiester bonds. Restriction enzymes are widely used in laboratories as a very important tool for DNA manipulation. It finds application in gene cloning techniques, mapping of single nucleotide polymorphisms, DNA fingerprinting, southern blot analysis, etc.
Ssp I restriction endonuclease by Genei cleaves the DNA at the palindromic sequence AAT/AAT. Due to the pattern of the cleavage, this restriction endonuclease is known to generate blunt ends. The enzyme is isolated from the bacterium Sphaerotilus natans.
